Ultimovacs kan se tillbaka på ett starkt 2020

2020 var ett år av viktiga kliniska framsteg för Ultimovacs. Det är speciellt noterbart att bolagets redan omfattande utvecklingsprogram med toppkandidaten UV1 breddades genom två ytterligare fas II-studier. BioStock pratade med vd Carlos de Sousa, som tog över vd-rollen halvvägs genom 2020, för att ta del av hans intryck av ett händelserikt år för Ultimovacs och vad han förväntar sig 2021.

Ultimovacs huvudmål är att utveckla ett universellt cancervaccin som kan fungera i kombination med andra behandlingar i syfte att stärka patientens immunförsvar och minska den totala cancerbördan. För att uppnå detta mål har bolaget inlett flera kliniska studier som testar deras immunterapiläkemedel UV1 över ett brett spektrum av cancerindikationer och i kombination med andra typer av behandlingar, särskilt checkpointhämmare (kontrollpunkthämmare).

Fas I studier

Det kliniska utvecklingsprogrammet med UV1 inkluderar kliniska prövningar på sjukhus i Europa och USA. Fyra av studierna befinner sig i fas I, tre har redan avslutat behandlingar och genomgår uppdateringar gällande överlevnadsdata. Av de tre studierna är en inom prostatacancer, en inom icke-småcellig lungcancer (NSCLC) och en inom metastaserande malignt melanom. Alla tre har genomförts i samarbete med Oslo universitetssjukhus. Den fjärde fas I-studien gäller även den malignt melanom, och är en pågående USA-baserad studie som testar UV1 i kombination med checkpointhämmaren pembrolizumab som första linjens behandling med 30 patienter. Studien är utformad för att bedöma UV1:s säkerhet och tolerabilitet samt för att undersöka initiala tecken på klinisk respons. I augusti tillkännagav Ultimovacs att patientregistreringen avslutats, och i september bekräftade bolaget att studien uppnått de primära målen för säkerhet och tolerans.

Fas II studier

I början av 2020 hade Ultimovacs två studier i fas II, INITIUM och NIPU. Båda är proof-of-concept-studier som testar trippelkombinationen av UV1 med checkpointhämmarna ipilimumab och nivolumab i metastaserande malignt melanom respektive mesoteliom. NIPU-studien, där den första patienten doserades den 15 juni 2020, genomförs i samarbete med Oslo universitetssjukhus och läkemedelsjätten Bristol-Myers Squibb (BMS).

Under 2020 tillkännagav Ultimovacs ytterligare två fas II-projekt. I maj meddelade bolaget ett fåtal detaljer gällande en studie som ska genomföras i samarbete med ett stort ledande läkemedelsbolag och är sponsrat av en klinisk prövningsgrupp. För bara några veckor sedan meddelade Ultimovacs att den stora läkemedelspartnern i DOVACC-studien är AstraZeneca och att Nordic Society of Gynecological Oncology (NSGO) och European Network of Gynecological Oncological Trial Groups (ENGOT) kommer att delta i studien. Studien ska utvärdera UV1 i kombination med AstraZenecas durvalumab, en PD-L1-checkpointshämmare och dess PARP-hämmare, olaparib, i BRCA-negativa patienter med höggradig avancerad äggstockscancer. Strax innan jul tillkännagav Ultimovacs att man inleder ytterligare en fas II-studie, den här gången ska UV1 testas i kombination med pembrolizumab hos patienter med huvud- och halscancer.  Studien, kallad FOCUS, kommer att genomföras på tio kliniker i Tyskland och leds av välrenommerade professor Mascha Binder. Starka finanser och starkt ledarskap

Samtidigt som mycket hände i Ultimovacs kliniska pipeline under 2020, tog bolaget också några viktiga steg för att stärka sin finansiella position och sin ledning. För att fortsätta stödja de de kliniska framstegen genomförde Ultimovacs en betydande private placement i maj riktad mot norska och internationella institutionella investerare. Kapitalanskaffningen tillförde bolaget cirka 160 MNOK (cirka 157 Mkr). I slutet av tredje kvartalet rapporterade Ultimovacs en total kassaposition på 453,5 MNOK (cirka 430 Mkr), vilket förväntas räcka till att ta bolaget genom resultatavläsning av primära slutpunkter i fas II-studierna 2022 och 2023.

För att stärka bolagets affärsutvecklingskapacitet när bolaget nu börjar närmar sig kommersialisering, utnämnde Ultimovacs Carlos de Sousa till vd i februari, och han började i sin nya roll i juni. De Sousa har mer än 30 års erfarenhet av entreprenörskap inom bioteknik -och läkemedelssektorn, både inom små nystartade bolag och big pharma.

Vd:n om 2020 och vad som väntar härnäst

BioStock kontaktade den nya vd:n för att höra hur han ser på Ultimovacs 2020 och vad vi kan vänta oss av bolaget under 2021.

Carlos de Sousa, what would you say is the main highlight of 2020 for Ultimovacs?

– It has been a busy but successful year on all fronts for Ultimovacs. However, if I had to point to one highlight in particular, I would say that it was our ability to setup four clinical phase II trials and begin patient enrolment in two of these. We expect a total of about 500 patients enrolled for these four studies.

Since taking the spot as CEO in June, what have been the most challenging and the most gratifying moments of your tenure in 2020?

– The most challenging moment has probably been joining Ultimovacs in the middle of a pandemic. Maintaining performance and engagement from the team to drive our plans forward in the most optimal way, at a time when most activities were done from a distance, was a major challenge.

– Despite this challenge, it has been gratifying to see that the team responded very well to my entrance as CEO. In fact, team spirit has not faltered and dedication to our projects has remained very high.

Ultimovacs has engaged in two new phase II trials during 2020, one of which is in partnership with AstraZeneca. How impactful are partnerships of this calibre for Ultimovacs?

– The partnership with AstraZeneca reinforces our collaboration with another big pharma, Brystol-Myers Squibb for our NIPU study. Partnerships with big pharma are very difficult to obtain, and when they happen, they are a validation of our idea and innovative strength, as well as our clinical approach, especially when it involves tough indications like mesothelioma or ovarian cancer in the NIPU and DOVACC trials, respectively.

– It is important to note that the same goes for partnerships with top clinical trial networks like ENGOT. ENGOT is one of the most respected oncology networks in Europe, specifically within Gynaecology. The collaboration with them indicates that the medical community is interested in UV1 and its potential.

– Furthermore, regarding our FOCUS trial, the fourth phase II trial announced last year, I should add that we have seen enormous interest from Key Opinion Leaders there as well.

What is the preliminary timeline for the DOVACC study?

– We expect the first patient to be treated during the first half of 2021, and topline data readout of the primary endpoint is expected for 2023.

2020 has been marred by a pandemic, which has, in many cases, caused several delays in clinical trials. Has Ultimovacs been affected directly, and do you expect any disruptions this year?

– Clinics everywhere have seen disruptions and Ultimovacs was not spared by the pandemic. Particularly in the beginning of the pandemic, we experienced a 2- 3-month delay in patient enrolment. However, I can proudly say that we have managed to continue delivering despite the delays and the challenges, and a lot credit goes to the team effort I mentioned before.

– Moving forward, a lot will depend on the extent of restrictions related to the pandemic as well as the vaccine rollout. The biggest challenge will continue to be with patient enrolment, as many cancer patients are, understandably, afraid of entering hospitals knowing that Covid is still going strong. However, we remain optimistic and confident that as more people get vaccinated, restrictions will soon subside, and patients will be more comfortable coming into the clinics.

 Finally, now that 2020 is behind us, what is your vision for the company in 2021?

– Our key objectives are to continue running the studies as smoothly as possible despite the constraints and making sure that we increase awareness of Ultimovacs and our approach throughout Europe as well as abroad in the US and in Asia. This will also go a long way to gaining potential investors and partners.

– Another objective will be to continue progressing with our TET platform. In our TENDU study with prostate cancer patients, the first patient is expected for enrolment in Q1 of this year.
